Introduction: The aim of our study was to examine the influence of recombinant human erythropoietin (rhEpo) on the brain infarct volume and neuronal degeneration in the cortex of rats exposed to the right middle cerebral artery occlusion (MCAO). Materials and methods: Focal cerebral ischemia was induced in male Hannover Wistar rats by right MCAO for 1 hr. Sham operated, vehicle treated animals served as the control group. Ischemic animals received either vehicle or rhEpo (5000 IU/kg, i.p.), 3 h after induction of ischemia. Rats were sacrificed 24 h after the start of ischemic or sham experimental procedure. Extent of the brain infarct volume was determined using the TTC dye at 24 h after onset of ischemic procedure in ischemic, vehicle treated animals such as in ischemic animals treated with rhEpo. In the mentioned experimental groups, Fluoro- Jade B histofluorescence was used for the evaluation of cortical neurons undergoing neurodegenerative changes and immunohistochemical detection of NeuN for neuronal loss measurement. Results: The results of our experiments showed that MCAO/reperfusion cause marked cortical infarction. Prominent Fluoro-Jade B staining and the decrease in NeuN immunoreactivity, following MCAO were also demonstrated. Administration of rhEpo significantly reduced the infarct volume, neuronal degeneration and neuronal loss in the ischemic animals in comparison to the rats of the control group. Conclusion: Our results clearly indicated neuroprotective potential of rhEpo administered after induction of MCAO.
