Optimization of ultrasonication assisted extraction of bioactive components from Medicago sativa: Influence of extraction solvent, temperature and pH

Medicago sativa L., Fabaceae (alfalfa or lucerne), is a perennial herbaceous plant species that has a long history of medicinal and nutritional use in Europe, Asia and America. It is assumed that its medicinal properties are mostly based on the content of phenolic compounds, among which isoflavones play an especially prominent role. Ultrasonic-assisted extraction (UAE), an inexpensive, rapid, simple and efficient technique, was used for extraction of isoflavone and other antioxidant compounds from aerial parts of Medicago sativa L., Fabaceae. The proce-dure was performed according to a three-level-three-factor Box–Behnken design. The inde-pendent variables were ethanol concentration (X1), temperature (X2), and extraction solvent pH (X3), while the responses were yield of the extraction, DPPH radical scavenging activity of the extracts (RSA IC50), content of total phenols (TP), total flavonoids (TF), total phenolic acids (TPA) and total isoflavonoids (TI). Concentration of isoflavones in the obtained extracts was determined using HPLC while the other responses were determined spectrophotometri-cally. The results were analysed using response surface methodology. By applying multiple regression analysis on the experimental data and by analysis of variance (ANOVA) for the selected models it was shown that the relationship between dependent and independent variables can best be described with quadratic models. In addition, each of the independent variables, as well as their interactions, had a significant (P < 0.05) effect on at least one of the selected responses. The response values of extracts prepared at optimum conditions (response, X1, X2, X3) were: yield (162 mg/ml, 20%, 60 oC, 5.5), TI (236.5 mg/ml, 0%, 35 oC, 8.5) TP (6.63 mg/ml, 100%, 60 oC, 5.5) TF (0.292 mg/ml, 100%, 23 oC, 5.5) TPA (0.275 mg/ml, 100%, 23 oC, 8.5) and RSA IC50 (3.97 mg/ml, 40%, 60 oC, 7.0). In addition, it was found that the most abundant isoflavones in M. sativa were glycosides of formononetin (5.7-206.1 ug/ml) and genistein (2.5-37.3 ug/ml). The predicted values matched well with the observed ones indicating the suitability of the models for extraction of bioactive compounds from M. sativa.
