Application of PET scan in detection of FDG distribution in chicken embryo
Detection of fluorodeoxyglucose (FDG) using positron emission tomography (PET scan) is widely used in diagnostic and research of brain, heart and neoplastic tissue metabolism. Small laboratory mammals (mice, rats) are widely used as a model in that field. Chicken embryos have been just recently used as a model as simple and convenient organisms for above mentioned research. But optimal way and dose of application is not yet defined. In this study 54 13-days-old embryos of ROSS hybrid were used and 12 MBq of FDG was applied i/v over the air chamber into chorioallantoic membrane (CAM) vessels. Immediately after application embryos were scanned using Raytest ClearPET high performance small-animal PET scanner (Rudjer Boskovic Institute, Croatia) for 1 hour. After the appropriate 2D and 3D image was obtained, the distribution of the FDG in different tissues was analysed. As in mammalian models, the main activity of FDG was detected in heart, brain and kidneys, but also in yolk sac, which is a very active organ in avian species. As some previous results showed, chicken embryos could be effectively used as a model for testing different radiopharmaceuticals, but also as a model for detection of dynamics of spread of different neoplastic tissue and its possible treatment and possible research of heart, kidney and brain disorders. This study showed that FDG could be effectively applied into embryos over the CAM vessels under the air chamber and that during one hour of scanning all important tissues could be visualized.
