Chemotaxonomic investigation of Globularia L. taxa
Fifteen Globularia samples were collected from different natural populations in Croatia during their flowering stages in 2015, including the following taxa: G. alypum, G. cordifolia (ssp. cordifolia and ssp. bellidifolia) and G. punctata. In order to evaluate the differences in their phytochemical profiles, methanolic extracts of aerial parts were prepared by 24-hour maceration. Afterwards, all extracts were subjected to thin- layer chromatography (TLC) and liquid chromatography-photodiode array detection- electrospray ionization-tandem mass spectrometry (LC-PDA-ESI-MSn) to separate and analyze individual components. Identification of compounds was enabled by the simultaneous use of reference standards, while the obtained mass fragmentation data, in combination with literature data, enabled the identification of compounds for which reference standards were not available. The results of this study confirmed the Globularia species to be characterized mainly by iridoid and phenylethanoid compounds, while more flavonoids were observed in the samples obtained from G. punctata populations. Both methods provided good differentiation between G. alypum (globularin, calceolarioside A, globularioside), the samples of G. cordifolia (globularifolin, 6'-O- benzoylmonomelittoside, asperuloside) and the samples of G. punctata populations (globularin, asperuloside, besperuloside, trichosanthoside A). The two taxa belonging to the G. cordifolia populations could not be unambiguously distinguished based on their observed chemical composition. However, one population contained only trace amounts of asperuloside, a major compound in all other investigated populations of G. cordifolia. The visually observed phytochemical differences were further evaluated using principal component analysis (PCA) and hierarchical cluster analysis (HCA) resulting in two main clusters, the G. cordifolia and the G. punctata cluster, together with two outliers (G. alypum and the low asperuloside-G. cordifolia).
