Carbohydrate Monomer Profiling of Waste Plant Biomass Using HPTLC and HPLC-PDA
In recent years waste plant biomass has become a perspective source for the production of a variety of bio-based products which rely on the recovery and reutilisation of their most dominant component, carbohydrates. The aim of this study was to evaluate the performance of HPTLC and HPLC-PDA with pre-column derivatization in determining monomeric carbohydrate composition of sugar beet pulp, walnut shell, cocoa bean husk, onion peel and pea pods. HPTLC was performed on Silica gel 60 plates and optimized with respect to plate impregnation, composition of the developing solvent and total length of plate development. HPLC-PDA method was conducted using 3-methyl-1-phenyl-2-pyrazoline-5-one pre-column labelling and separation on standard C18 column under optimized conditions. Identified monomeric components using both methods comprised 6 neutral sugars and 1 uronic acid. HPTLC monomeric profiles exhibited glucose as dominant component in all samples, ranging from 74.7% (onion peel) to 80.4% (walnut shell) of all sugar content. Markedly lower glucose results obtained by HPLC-PDA indicated a notable influence of the method used. Upon setting sugar beet pulp as a reference, due to its previously well-described composition, the observed difference was considered to result from the underestimation of minorly represented sugar components in HPTLC, consequently reflecting in the overestimation of glucose relative content by 38, 4% (walnut shell) to 50, 3% (onion peel). Impaired quantification precision was ascribed to the increased diffusivity of sugar analytes in HPTLC system. However, the employed HPTLC exhibits potential for rapid qualitative analysis of sugar composition of complex substrates, while HPLC-PDA as preferred for precise quantification.
