Outbreak of bovine tuberculosis in a beef cattle herd in Slovenia
Since 2009, Slovenia has been offi cially free of bovine tuberculosis (bTB) and there had previously been only one outbreak in newly imported animals in last 20 years. However, in 2015 visible tuberculous lesions were detected at regular slaughter of three young beef bulls. In all three animals, M. caprae was confi rmed by bacteriological culture of tissues. All animals were from the same herd of beef cattle. Nine of the remaining 19 animals showed a positive or suspicious reaction to comparative intradermal tuberculin test and all were culled. In 6 of them, the infection with M. caprae was confirmed by bacteriological examination. First three bulls were imported from Austria at age 3 months and were slaughtered at age 22 months. In the meantime, the animals from Romania, Germany and Czech Republic were introduced into the herd. One strain of M. caprae from each positive group was genotyped using variable number tandem repeat targeted at genetic loci that contain variable numbers of repeated sequences (MIRU VNTR method, 22 loci). Band sizes from MIRU- VNTR-22 results were translated into numbers of individual repeats and results were presented in the form of 22-digit numerical codes. Codes were analysed using the categorical coefficient and unweighted pair group method with arithmetic mean (UPGMA). Isolates was compared to those deposited in database of Croatian National Reference Laboratory collected in period from 2009-2015. Interestingly, two different genotypes from the same outbreak have been identified. One type was close to the Central European isolates and the other to the Eastern European isolates. The results show that trading of cattle within EU allows spreading of bTB.
