Changes in BV-2 microglia susceptibility to murine cyotmegalovirus as a result of varying cultivation conditions

Murine microglial BV-2 cell line is commonly used as a substitute for primary microglia. They are commonly cultivated in the Dulbecco’s modified eagle medium (DMEM) supplemented with fetal calf serum (FCS) and L-glutamine (L-G). This cellular environment is unable to imitate the one present in the central nervous system, thus causing morphological and functional changes to the microglial cells. Microglial cells are reactive by nature, and as such are affected by all changes in their environment. BV-2 microglia cells exhibited low susceptibility to murine cytomegalovirus (MCMV) in the first experiments we performed with the goal of establishing an in-vitro infection model. As a result, our aim was to determine whether the changes in the condition of cultivation can affect the susceptibility of BV-2 cells to MCMV infection. For the purposes of these experiments cells were grown in the FCS –depleted cultivation medium with or without the presence of L-G. Furthermore, to achieve more natural micro enviromental conditions ATP and tocopherol-α in various concentrations were added as a substitute. Results from our study show that manipulation with the cultivation conditions specifically FCS depletion, leads to morphological changes of BV-2 microglia cells. Complete FCS depletion resulted in increased susceptibility of this cell line for MCMV-infection. Varying FCS depletion also increased pro-inflammatory markers CD86 and iNOS, depending on the concentration. This indicates that the environmental changes that modulate the metabolic and immunological profile can cause an altered response of microglial cells to viral pathogens.
