Variability of major histocompatibility complex class II DRB1 locus in wild boar (Sus scrofa l.) from Croatia – preliminary observations

Major histocompatibility complex contains some of the most variable genes in vertebrates whose diversity has been driven by evolutionary processes over long period of time. This large region of DNA through class I and II genes encodes cell-surface proteins that bind antigens derived from pathogens or parasites and present them to T-lymphocytes, thus playing a major role in immune response. Knowing wild boar's widespread distribution and potential to host many parasite species, makes it suitable for testing correlation between MHC haplotypes and susceptibility/resistance to diseases. The goal of this study was to investigate imunogenetic variability of major histocompatibility complex class II DRB1 locus in Croatian wild boars. We analysed 73 samples of muscle tissue of wild boars from whole Croatia, excluding region that belongs to Zagreb City. Samples were collected following regular hunting operations, cut to appropriate size, stored in 96% alcohol, signed and frozen. DNA was extracted using commercial kit. Polymerase chain reactions were used to amplify the exon 2 of DRB1 locus and successfully amplified sequences were sequenced. The exon 2 was used as an indicator of allelic diversity since it encodes the peptide-binding region of MHC receptor and is the site of most polymorphism. Suitable bioinformatics programs (BioEdit, SeqScape) were used for analyses. We identified 11 different DRB1 alleles in wild boars in Croatia, with allele SLA‐DRB1*02 being the most frequent. However, only four animals were heterozygous (5.5%), suggesting existence of null-alleles, which will be resolved in further study. The results of this preliminary study will be used for future research of correlation between the MHC haplotypes and specific parasitic diseases of wild boar.
