Adhesion properties of probiotic S-layer-expressing Lactobacillus strains

S-layer proteins form the outermost layer of bacteria, which could be involved in adhesion and as such, could have the potential to enhance the inhibition of binding of pathogenic microorganisms, which is known as competitive exclusion. For this reason, adhesion of four Lactobacillus strains – Lactobacillus brevis D6, L. brevis ZG1, L. paraplantarum SF9B and L. brevis SF15B, which express S-layer proteins, were investigated using in vitro adhesion models. All the experiments were performed using Lactobacillus cells of certain strain before and after removal of S-layer proteins from their surface, in order to examine the role of S-layers in adhesion process. Surface proteins involved in adhesion could adhere either directly to the epithelium or to specific components of the epithelial environment. The extracellular matrix surrounding mammalian epithelial cells is composed of various proteins, including laminin, collagen and fibronectin. Hence, in vitro binding of Lactobacillus cells to named ECM proteins was studied, as well as to mucin which is the main component of mucus layer in the colon and in the small intestine. Moreover, adhesion to cell line of Caco-2, human epithelial colorectal adenocarcinoma cells was examined, while the same cell line was used in experiments of competitive exclusion. Cells of all the tested strains adhered to Caco-2 cell lines more effectively and with higher affinity to mucin with present S-layer proteins. The poor, but not completely abolished adhesiveness of the S-layer-depleted Lactobacillus cells to ECM proteins is in accordance with the hypothesis that S-layer subunits indeed are adhesion proteins ; however, there are some other factors included in adhesion process of the bacterial cells. Additionally, presence of Lactobacillus cells caused decrease in adhesion of S. Typhimurium FP1 and E. coli 3014 to cells of Caco-2 cell line. All the obtained results indicate that S-layer proteins ensure better adhesion ability of probiotic cells and that S-layer-expressing strains could be more effective in competitive exclusion of pathogens.
