The effect of various levels of fluoride (amine fluoride) on the KOH-soluble and structurally bound fluoride in human enamel:an in vitro study

The concentrations of fluoride in agents for professional application are empirically chosen and not based on dose/response studies. The aim of this study was to investigate the relationship between different fluoride concentrations and the acquisition of KOH-soluble and structurally bound fluoride in human dental enamel in vitro. Four slabs were prepared from each of 31 impacted, extracted third molars and selected at random so that one slab from each tooth was present in each group. The slabs of three groups were immersed for 3 min. in an amine fluoride solution: (A) 1% F ; (B) 0.5% F ; (C) 0.25% F during three days. One group of slabs was not treated and served as the control (D). The amounts of KOH-soluble fluoride (ľg/cm2 ą SD) were: (A) 35.2 ą 16.1 ; (B) 33.2 ą 16.3 ; (C) 18.6 ą 9.2 ; A and B did not differ significantly, and were superior to C (analysis variance LSD at p=0.05). The enamel slabs of all treated groups took up considerable amounts of structurally bound fluoride (p<0.05). Fluoride uptake values were (ppm F ą SD): (A) 1280.6 ą 1178.4 ; (B) 986.9 ą 542.7 ; (C) 809.6 ą 861.6 and in the control group (D) 275.3 ą 95.3. The differences in the amounts of structurally acquired fluoride were significant between the A and C groups. The results showed that lowering the standard fluoride concentration of amine fluoride solution (1% F) by half did not reduce the amounts of KOH-soluble and structurally bound fluoride. It can be stated that if difference in the efficacy of the standard and half strength amine fluoride solutions exists, it is probably of little clinical significance.
