Sažetak
Thyroid cancer is one of the fastest growing types of cancer in the world with annual percent change between 0.8 and 24.2. Presented results are part of three-phase-study that aims to improve cancer prevention. In the phase one of the study, the genome integrity is biomonitored using battery of tests in patients with thyroid diseases. Phase two was done in order to determine impact of oxidative stress on the levels of DNA damage. Finally, in the phase three, evaluation of expression of variant B-Raf and Ret proteins was done. The study population consisted of 83 volunteer patients (70 female:13 male, average age 52.00±12.63 years, 22 smokers) diagnosed with follicular adenoma (33), papillary cancer (26), and other thyroid diseases (goitre and thyroiditis, 24). Compared to healthy population consisted of 83 volunteers (matched for gender, age, and smoking status) the population with thyroid diseases showed 1.51- fold higher number of chromosome aberrations, 1.78-fold higher percentage of DNA in comet’s tails, and 2.35-fold higher number of micronuclei. Furthermore, malondialdehyde as the parameter of lipid peroxidation was assessed and 1.65-fold higher blood concentration in the patients’ group was detected. On the contrary the levels of glutathione were 1.49-fold higher in the control group. Finally, 82.50% of thyroid tissues from diseased volunteers showed positive expression of B-Raf, while only 21.25% of thyroid tissues was positive for Ret protein. The results of this study suggest that patients with thyroid diseases have more DNA damage and worse oxidative status. Overall results will provide great basis for detection of biomarkers that could be used for better risk assessment of cancer diseases and therefore for improvement of cancer prevention.
