ABC transporters in antibiotic clusters
ATP-binding cassette (ABC) proteins utilize ATP to act as uptake or efflux transporters. ABC protein superfamily is one of the largest groups of proteins, found in all organisms. Among numerous other functions, they can be involved in the secretion of antibiotics through the cell membrane. Therefore, they contribute significantly to self-resistance of the producing organisms, but also of other species, including important pathogens. Identification of antibiotic efflux- transporters can help in the development of more efficient antibiotic production systems. Also, design of specific inhibitors of antibiotic efflux is important for the improved control of multidrug-resistant strains of bacteria. The aim of our work was to collect and classify ABC transporters that are present in antibiotic- producing gene clusters. We have retrieved 320 previously described antibiotic gene clusters from diverse prokaryotic and eukaryotic microorganisms and found that 31.9 % of them contained at least one ABC transporter sequence. Out of 74 Pfam families belonging to the ABC superfamily according to the Transporter Classification Database (TCDB), we have found 29 in antibiotic clusters. Interestingly, only 1 ABC transporter sequence was found in eukaryotic (fungal) antibiotic-producing clusters. We will present preliminary results of clustering and phylogenetic analysis of the retrieved 246 ABC transporters sequences and compare the identified groups regarding their presumed physiological role and transport mechanism. Overall, our results will provide a systematization of knowledge on ABC transporters that are present in antibiotic- producing gene clusters, and easy implementation to newly identified transporters.
