Distribution of silver in organs of Wistar rats after chronic exposure to silver nanoparticles
The rapid progress and early commercial acceptance of silver-based nanomaterials, owed to their biocidal activity, urges a need for assessment of the potential adverse health effects of nanoparticles due to prolonged exposure. Oral toxicity of silver nanoparticles (AgNPs) is of particular concern to ensure public and consumer health due to the widespread use of AgNPs in products related to food and food contact materials. We investigated tissue distribution of silver in different organs of Wistar rats after 28-day oral exposure to polyviniylpyrrolidone-coated AgNPs (Ag= 0.1 and 1 mg/kg body weight). The organ distribution pattern of silver following administration of AgNPs was determined with ICPMS after microwave assisted acid digestion. Silver was present in all examined organs with the highest levels in the coecum and colon in both sexes. In males, besides the intestinal system, the highest silver concentrations were detected in kidney, brain and epididymis, followed by liver, brain and ovary. The lowest silver content was found in heart, duodenum and ileum in both sexes. The consequences of in vivo retention of silver in brain, kidney and sex glands should be considered in a risk assessment of AgNPs.
