Expression profiling of obesity/adipogenesis- related genes in rats with altered serotonergic homeostasis

Serotonin (5-hydroxytryptamine, 5HT) has been implicated in mechanisms that control body weight and, if dysfunctional, predispose to obesity. Reduced brain 5HT activity is generally associated with the increase in food intake and body weight ; however, exact molecular pathways involved in serotonergic modulation of energy balance have yet to be fully understood. The aim of the present study was to identify molecular candidates mediating serotonergic regulation of energy balance, by profiling the expression levels of obesity/adipogenesis-related genes in hypothalamus of rats with altered serotonergic homeostasis (Wistar Zagreb-5HT rats, WZ-5HT rats). High-5HT (hyperserotonergic) and low-5HT (hyposerotonergic) sublines of WZ-5HT rats were developed by selective breeding towards extreme activities of platelet serotonin transporter. In addition to differences in peripheral and central serotonin parameters, animals from the two sublines display clear differences in the lifelong body weight. Our gene expression profiling, using PCR array technology, has indicated differential expression of multiple obesity- and adipogenesis-related genes in hypothalamic cDNA pools generated from animals of each subline. Interrelation of these genes will be further investigated using quantitative real-time RT-PCR and ELISA measurements in samples from individual animals.
