Talens and CRISPR/Cas9: Generation of genome engineered mouse models using editable nucleases: Practical experience.
Targeting strategies based on programmable nucleases are a rapid and efficient system for site-specific knock-in and knock-out of gene of interest. These nucleases include zinc-finger nucleases (ZFN), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short palindromic repeats (CRISPR)/Cas RNA-guided nucleases. Therefore, the programmable nucleases are a method of choice for creation of mouse models where precision and speed is required. This presentation gives several practical examples of mouse models generated at the Czech Centre for Phenogenomics.
