A Multidisciplinary Approach to the Study of Adamantyl Aminoguanidines, a New Class of Potentially Bioactive Compounds
Adamantane is a common structural motif in a number of bioactive compounds due to its unique properties such as high lipophilicity and chemical inertness [1]. Adamantane-containing derivatives have been used in medicinal chemistry for more than 50 years and the discovery of new polycyclic drugs continues up to the present day [2]. The goal of this study was to combine the lipophilic adamantyl fragment with a highly polar guanidine moiety in order to see how that kind of a system would work and to search for new properties and possible applications of such molecules. The intention was also to add variety in the type and the nature of the bond connecting these two systems and therefore we used the aminoguanidine subunit. The resulting adamantyl aminoguanidines are compounds with a high potential of use as pharmacophores but their preparation and characterization has only recently been described [3]. Here a detailed study of this class of compounds is presented, with special emphasis on their physico-chemical properties as well as the evaluation of the possibilities of their biological application.
