In vitro protocols for the evaluation of biocompatibility of metallic nanoparticles
Due to the increasing use of metallic nanoparticles (NPs) in a wide variety of products, there is urgent need for safety evaluation and toxicological studies of these NPs. However, popular properties of metal-based NP such as catalytic reactivity, high surface area, light emission properties, have the potential to interfere in many well-established testing protocols. Cell viability assay are among the most versatile and popular assays used for in vitro toxicology. We outline the most frequently used cell viability assays to assess the cytotoxicity and biological reactivity of two types of metallic NPs: silver and maghemite. We identified key issues that need to be addressed in relation to inclusion of relevant controls, assessing particles for their ability to interfere in the assays, and using systematic approaches to prevent misinterpretation of data. The aim of this presentation is to share knowledge relating to NPs toxicity testing in order to provide advice and support for guidelines, regulatory bodies, and for scientists in general.
