In-line monitoring of API synthesis and product characterisation
In recent years, process analytical technology (PAT) has emerged as one of the key elements to fully implement in-line analytical techniques for process monitoring. In-line techniques based on vibrational spectroscopy enable in situ monitoring of physical and chemical transformations and provide insight into the mixture composition at a given time. Here we report on using in-line Raman spectroscopy for monitoring the synthesis of an active pharmaceutical ingredient (API) shown in Figure 1. This API is used as a selective proton pump inhibitor. Raman spectroscopy was employed for the remote detection through a fibre optic probe which facilitated real-time monitoring of the API synthesis. In order to unambiguously identify the reaction product, the obtained crystals were further evaluated off-line using NMR spectroscopy in solution and solid state.
