Aiolos transcription factor activity in immature and malignant human lymphocytes
Studies of lymphoid-specific gene expression have identified a number of transcription factors as putative controllers of T and B lymphoyte differentiation and maturation. It has been shown that mice homozygous for a deletion in the Aiolos gene undrgo dramatic changes in their B cell populations. Moreover, aging animals with the same mutation develop lymphoproliferative disorders, mainly B cell lymhomas. Given the imoportance of the Aiolos gene in the mouse lymphopoietic system we wanted to investigae if an Aiolos homologue expressed in human lymphocytes has a role in human lymohomas. We have successfully amplified human mRNA using primers for the Aiolos cDNA. Also, we have adjusted and established a method for specific conditions of RNA isolation from formalin fixed paraffin embedded tissues. Hence we were able to analyze Aiolos mRNA from archive tissue specimens from 30 patients with Hodgkin's and non-Hodgkin's lymphoma. We have shown Aiolos gene expression in all but one specimen of the lymphoma tested. We also detected Aiolos amplification products of different size indicating that at least in humans there might be Aiolos spicing vriants.
