Interaction of 4-Hydroxynonenal and Albumin
4-Hydroxynonenal (HNE) is a second toxic messenger of oxygen free radicals, which can be formed by oxidation of polyunsaturated fatty acids in complex chain reactions in vivo and in vitro. It is physiologically present in organism as free aldehyde as well as bound to the macromolecules. In high concentrations (100 uM) HNE can cause cell death in vitro, but in lower concentrations, which are not cytotoxic (less than 10 uM), it can influence cell proliferation and differentiation. Due to high reactivity of the aldehyde, almost any molecule in its surrounding can be modified by HNE. Proteins are common target for modifications by HNE while aldehyde can react with thiol and amino groups present in protein. Such modification can change protein structure and consequently it can influence protein function. In case of growth factors or enzymes, even minor modifications in proper position can cause protein disfunction not affecting total protein structure, but changing binding site for a growth factor or active site of an enzyme. In our work we used in vitro system to investigate interaction of proteins and HNE, based on bovine serum albumin (BSA) as a protein model and spectofluorometical and immunochemical methods to determine sensitivity of the methods to detect changes in albumin molecule caused by HNE. As free HNE can be detected by UV detection at 220-223 nm where it shows strong absorption maximum, changes in HNE spectra were also recorded.
