Sažetak
Metallothioneins (MTs) are low molecular mass proteins, which have characteristic metal-binding properties because of thier high cysteine content. MT content was determined using method introduced by Brdička (1933). It is method for polarographic determination of proteins which contain thiol groups, and it is performed in ammonia solution of cobalt(III) salt. Method was modified by Paleček and Pechan (1971), and Olafson and Sim (1979). Determination of proteins which conatin thiol groups (including metallothioneins) is based on lineas relationship between catalytic hydrogen wave and the protein concentration, and a method of differential pulse voltammetry with previously determined calibration curve was applied. Since this method is very sensitive and the linearity range is very narrow, special care has to be taken in a way that measured values in the inknown sample have to be in the range of the calibration curve. In samples isolated from mussel digestive gland exposed to high concentrations of cadmium, high contents of MT were observed, and the measured values are often above the calibration range. The results of MT measurement in the samples which were diluted 10 times during their preparation (before the thermal treatment of the samples) and in the samples diluted immediately before measurement, will be presented. Statistical procedure of the measured data was performed using standard statistical tests using Excel computer software.
