Combination of biomaterials and stem cells for brain transplantation after ischemic injury in mouse
Neural stem cells (NSC) were incorporated in the soft alginate hydrogel beads and tested in vitro prior to transplantation and in vivo after transplantation. NSC were viable 9 days after encapsulation witnin alginate beads and their number increased with time. Their differentiation to make neurons was demonstrated by immunocytochemistry with Map2 marker. The cells were marked with PKH26 and the alginate hydrogel-seeded NSC population was demonstrated in the brain 1 and 3 days after transplantation. Alginate hydrogels was shown to be biocompatible with stem cells and their transplantation. It can serve as an example of biomaterial to provide support to NSCs in the lesion-site directed stem cells therapy after stroke.
