Development of hyperbaric oxygenation protocol for the treatment of DSS-induced colitis in BALB/c mice
AIM: The aim of this study was to adjust the protocol of hyperbaric oxygenation (HBO2) for the treatment of DSS-induced colitis. METHODS: BALB/c mice at the age of 10-12 weeks were randomized into 5 groups: control mice (CTR), control mice undergoing HBO2 (CTR+HBO2), DSS treated mice (DSS), DSS treated mice undergoing 1x/day/2h/2.0bar and 2x/day/1h/2.3 of HBO2 (DSS+HBO2-1 and DSS+HBO2-2, respectively). Mice were fed 5% of DSS in drinking water ad libidum for 7 consecutive days. HBOT was administrated from day 1 to day 9. Disease activity (DAI) was assessed by daily measurement of mouse weight, stool consistency and the presence of blood per rectum. RESULTS: All DSS treated groups had significantly higher weight loss (WL) compared to both, CTR and CTR+HBO2 groups (p≤0.001). 2x/day/1h/2.3bar of HBO2 significantly reduced WL, which together with decreased DAI suggests improved disease outcome. There was no difference in WL in DSS+HBO2-1 mice compared to DSS group. Initial weight of same age female mice was significantly lower compared to male mice. CONCLUSION: Results of this study revealed importance of initial body weight for the sensitivity to DSS. Thus to reach better reliability and reproducibility of the data it is advisable to use mice of both sexes with body weight higher than 22g, irrespectively of age. In addition, two separate exposures to HBO2 had better healing effects than a single exposure.
