Sublines of rats with altered serotonin homeostasis
As a part of our (patho)physiological studies on peripheral and central serotonin (5-hydroxytryptamine, 5HT) compartments we have studied genetic mechanisms underlying the amine homeostasis in rats. By the breeding selection for the extreme values of platelet serotonin level (PSL) two sublines of Wistar-derived rats have been developed demonstrating significant differences in both peripheral and central 5HT parameters. Besides changes of various physiological/constitutional 5HT measures, the mentioned sublines displayed differences in the pharmacodynamic responses to the various 5HT related drugs. Pharmacogenetic differences studied as the response of platelet 5HT level to the administration of serotonin, its precursors, inhibitors of transmembrane serotonin transport and of vesicular serotonin storage will be reported. Moreover, pronounced differences in the extracellular 5HT levels in brain microdialysates of animals from the mentioned sublines, after the administration of specific serotonin blocker, will be presented. At the end a new selection paradigm consisting of directed breeding for the extremes of platelet serotonin transporter velocity in rats will be outlined.
