Sažetak
Growing any kind of cells in laboratory conditions brings the possibility of contamination. This could be prevented by addition of antibiotics and antimycotics to the culturing medium. However, the most authors agree that there are harmful effects of antibiotics and antimycotics. These effects include a wide variety of influences on cells from negligible to serious interference with cell proteomics. To measure such effects on cell proliferation of horse limbal stem cells we used flow cytometry. We tried to determine proportion of cells in various phases of mitotic cycle following three different treatments during culturing. We compared cells from primary culture with the cells treated with antibiotic-antimycotic (ABAM) and cells in the first passage. The cells with increased amount of ABAM had significantly lower values of the mitotic activity, as compared to the control cells from primary culture, but did not differ significantly when compared to cells in the first passage. These similarities in growth patterns of the cells increased amounts of ABAM and the cells form the first passage could be indicative of effect of ABAM. Because the cells in the first passage were still suitable for producing grafts, we assume that increased ABAM is required and useful treatment of stem cell transplant growth under conditions of increased risk of contamination
