Olive Oil Polyphenols Preconditioning Improves Liver Regeneration Induced By Partial Hepatectomy In Mice
Partial hepatectomy (pH) is a surgical procedure that stimulates a process of liver self-repair known as liver regeneration (LR). During this process hepatocytes are exposed to the oxidative stress associated with the metabolic overload which accounts for early events that contribute to the onset of LR. Polyphenols, independent of their antioxidant capacity, may act as hormetic effectors which affect gene signatures involved in preserving cellular homeostasis during stressful conditions. Hence, we investigated whether the preconditioning with olive oil polyphenols extract (PFE) may affect the development of LR induced by pH in mice via the stress response gene-profiles. Prior to pH, mice were administered PFE (50 mg/kg bwt ; i.p.) or saline during seven consecutive days while respective controls received vehicle alone. Hepatic oxidative state, involving redox state- regulating enzymes, thiol proteins, lipid peroxidation level and NFκB gene signatures was determined at different time intervals after pH. Liver mass restoration was calculated to assess hepatic regeneration. At the start of LR, PFE pre- treatment provoked transient increase in metabolic load, as indicated by the increased level of lipid peroxidation, depleted glutathione level and impaired processes that effect glutathione homeostasis, involving decreased glutathione reductase activity, suppressed γ-glutamylcysteine synthetase (GCS) transcriptional activity and increased glutathione utilization via the glutathione S-transferase mediated reaction. These changes were accompanied by the adaptive stress response induction, as indicated by NFκB, SOD2 and GCS gene upregulation, leading to the normalization of parameters of oxidative stress, recovery of glutathione status and ultimately resulting in increased liver mass recovery. The increase in oxidant and electrophilic load induced by PFE pre-treatment could play a role in the development of LR by sensitizing cells to acquire proliferative competence through the induction of NFκB-responsive genes.
