Sažetak
Individual differences in DNA damage response and lipid peroxidation level after exposure to ionising radiation (IR) are also due to single nucleotide polymorphism (SNP) in genes involved in DNA repair pathways. In order to estimate the influence of SNP on malondyaldehyde (MDA), as a product of lipid peroxidation, 58 individuals professionally exposed to low doses of IR were genotyped for hOGG1 (8-oxoguanined DNA glycosylase, Ser 326Cys) and XRCC1 (x-ray repair cross-complementing protein-group 1, Arg194Trp). Blood samples were irradiated with 2 Gy to 4 Gy (CO60). One or two polymorphic alleles were combined in one group (POL). T-test showed no significance between groups before and after radiation in MDA (Mean ± S.E.): (7.30±0.39), (7.36±0.46), 7.02±0.38) μmol L-1. After comparison before, and after 2 Gy and 4 Gy, results were XRCC1 POL: 7.02±0.47), (7.20±0.52), (7.04±0.47) μmol L-1 ; XRCC1 HO: (7.74±0.70), (7.61±0.87), (6.99±0.65) μmol L-1) ; hOGG1 POL: (7.80±0.53), (7.1±0.70), (6.30±0.56) μmol L-1 ; hOGG1 HO: 6, 90±0.57), (7.58±0.63), (7.61±0.50) μmol L-1. We found significant correlation among XRCC1 POL before and after both doses, while for XRCC1 HO there was correlation between the results before and after 2 Gy, and between 2 Gy and 4 Gy. HOGG1 POL correlated before and after 4 Gy ; and between the results before and after 2 Gy, and between 2 in all three groups. HOGG1 POL correlated before and after 4 Gy ; and between 2 Gy and 4 Gy radiation. hOGG1 HO correlated in all three groups. After t-test and Mann Whitney U-test comparison, there was a difference between the groups but it was not statistically significant. ANOVA showed an almost significant difference for hOGG1 groups (p=0.069), but not for XRCC1. Our conclusion is that the group was simply too small to estimate the real risk of exposure and conclude whether MDA is or is not a good biomarker for estimating occupational exposure to IR.
