Pharmacokinetics of salicylic acid in normal and tumor bearing mice

Malignant tumors affect normal host metabolism and hormonal balance. The changes are often described, although not well understood, and it seems that whole body processes become adapted to the tumor metabolic demands. Just as metabolism of normal nutrients, drug metabolism can also be changed in tumor bearing organisms. So, purpose of our work was to see if there are some difference in salicylic acid plasma level between healthy and tumor bearing mice. Female C57Bl6 mice were injected with F-10 melanoma cells into the hind limb and 15 days later experiment was performed. Animals were injected i.p. with acetylsalicylic acid (ASA), 20 mg/kg of body weight. Samples of blood were taken one hour or two hours after ASA was given and heparinized plasma samples were collected. Salicylic acid was extracted from plasma samples and determined by HPLC method with UV-detection. One hour after ASA was given, values of salicylic acid in plasma samples were the same in healthy and in tumor bearing mice (p>0.2). However, after two hours, salicylic acid values in plasma were lower in mice with tumor (p<0.01). Values of salicylic acid in plasma of healthy animals were equal one hour and two hours after ASA injection, while in tumor bearing mice, values after two hours were lower than those determined one hour after ASA (p<0.04). Although measured only at two time points, it is obvious that salicylic acid pharmacokinetics in healthy and in tumor bearing mice was different. Elimination of salicylic acid from blood was faster in mice with tumor than in healthy mice. Since only salicylic acid was measured, but not its metabolites, it remains to be proved if such differences are present due to the changes in the salicylic acid metabolism or its differential "distribution" in the organism.
