Establishing the baseline of physiological uptake of 18FDG in the myocardium in male rats
Positron emission tomography (PET) is a nuclear medicine imaging method used in the diagnosis and research of degenerative disease, tumors and heart and brain ischemic injuries. Tissue glucose uptake can be traced indirectly with PET using the glucose analogue - 18FDG as the radiopharmaceutical. Observation of the glucose uptake in the myocardium is used in cardiology for detecting viable myocardium after myocardial infarction or detecting ischemic myocardium parts in patients with coronary artery disease. Knowledge of the physiological glucose uptake is crucial for understanding, for proper interpretation and research of pathologically altered myocardium. Eight healthy, 8 weeks old, Wistar male rats were included in this study. 18FDG was injected i/v one hour prior to the scanning. Rats have been scanned on high performance small animal PET three times in 7 days intervals. Quantification of uptake was expressed by standardized uptake value (SUV). Only the left ventricle (LV) is regulary visible in the PET images, while the right one is visible only in cases of high SUV. In the anterior part of LV myocardium uptake is strongest, while in the apical is weakest. The uptake in posterior intraventricular sulcus region is of medium intensity and extends laterally. In the region of anterior interventricular sulcus, the distinctive ribbon pattern of lower uptake is often observed. Average SUV in first run was 3.89, in second 3.13 and in third 3.36. In successive scanning, spatial distribution of glucose uptake in the myocardium of each animal was very similar, while SUV has shown low to moderate variability. Despite the physiological tendency for variability in glucose uptake in the myocardium, the animals kept in controlled conditions and under the same PETscan protocol have shown relatively uniform glucose uptake into the myocardium, showing a credible and reproducible research environment for physiological uptake and diseases that affect glucose uptake in the myocardium.
