The effects og guanylin peptides on the bradykinin signaling
Atrial natriuretic peptide and urodilatin as agonists of GC-A inhibit bradykinin signaling pathway, while agonists of GC-B are not able to do so. In this study we would like to determine if agonists of GC-C, guanylin peptides, are able to inhibit bradykinin signaling pathway. We measured membrane potential of HEK-293 cells using whole cell patch clamp technique. Guanylin depolarized HEK 293 cells for 1, 5 mV (n=6), uroguanylin for 2, 1 mV (n=4) as well as their second messenger cGMP for 2.5 mV. All presented effects were inhibited by pottasium channel inhibitor, BaCl2 suggesting that those depolarizations are due to inhibition of pottasium channels. Bradykinin depolarized cells for 2 mV (n=11). When guanylin and uroguanylin are applied, depolarizations caused by bradykinin were inhibited. Those effects of guanylin peptides on bradykinin signaling pathway could be abolished by inhibition of regulator of G protein signaling protein using it's specific inhibitor. Those results are suggesting that interaction between agonists of GC-C and bradykinin signaling pathway is similar to one showed for agonists of GC-A and existence of an endogenous antagonist of the bradykinin signaling pathway.
