Molecular cloning and expression analysis of tumor necrosis factor alpha (TNFα) in reared Atlantic bluefin tuna (Thunnus thynnus)

Intensive culture of Atlantic bluefin tuna (Thunnus thynnus) BFT is limited by infectious diseases that, beside the stress, cause heavy losses. However, to date there are no reports of cloning and expression analysis of any major immunity gene of BFT, therefore the objectives of our study were to: report the full-length sequence of an cytokine with a major role in innate immunity, TNFα ; develop a 3D model for BFT TNFα protein ; measure in vivo tissue specific expression of TNFα in reared BFT during two years of farming process in order to evaluate gene importance as a health biomarker in tuna aquaculture. Cloning and sequencing of BFT TNFα using 5’and 3’ RACE gave full-length 1386bp long mRNA and 1889 bp long gDNA sequence, whose structures proved that it is a membrane-bound protein, which like other cytokines, contains AU-rich elements within the 3’ UTR. Phylogenetic and protein analysis showed that BFT TNFα has closest relationship with members of the Perciformes TNFα group. A 3D model for BFT TNFα was presented as a result of homology modelling using human TNFα as template. Analysis of BFT TNFα in vivo tissue specific expression evidenced that its expression level is reflecting health condition of reared fish therefore should be considered a potential health biomarker in tuna aquaculture. Our study also evidenced that liver being the site of enhanced TNFα production during systemic inflammation, should be considered as an important organ involved in fish defences mechanisms.
