GENOME STABILITY OF ZEBRAFISH (DANIO RERIO HAMILTON, 1822) AFTER EXPOSURE TO MODEL GENOTOXICANTS
DNA damage detected by Comet assay is not always a reliable indicator of genotoxic influence on genome integrity of exposed organisms. In order to evaluate DNA damage, its reparability and persistence that may lead to the alterations of DNA structure upon the genotoxic stress, in vivo and in vitro zebrafish models (cell line, larvae and adults) were exposed to model genotoxic agents. Persistence of DNA alterations after DNA damage detection and quantification by Comet assay was assessed by qRAPD (quantitative random amplified polymorphic DNA), AFLP (amplified fragment length polymorphism) and mitochondrial DNA control region sequencing. Furthermore, analysis of DNA repair and oxidative stress genes expression was performed as well. QRAPD and AFLP were able to detect DNA alterations even in cases when Comet assay indicated lack of genotoxic effect. Since such alterations can impair DNA structure and function, Comet assay results should be interpreted as an indication of DNA damage and preferably supplemented with other methods in order to predict the consequences of genotoxic insult more accurately.
