The Role of Pentadecapeptide BPC 157 in angiogenesis and NO-synthesis

A clear protection of G-I tract and an evident anti-inflammatory effect were shown for a novel stomach pentadecapeptide BPC(i.p./i.g.) in comparison with several reference standards in various ulcer models alog with a protection of endothelium. Thus, in the first experiments we evaluated whether this pentadecapeptide and other gastroprotective agents could affect the angiogenesis and healing process in vivo (using a slightly modified method of Szabo et al., AJM 9l, 1558, 1991). Number of new formed endothelial spacec inside new-formed granulation tissued in controls increased from 7, 94+/-1, 23 (day 3) to 14, 8+/-3, 12 (day 7) (P<0, 001). Significantly higher values were obtained in groups treated with pentadecapeptide (BPC 157) (solution 50 ug, 10 ug and 10 ng/ml) or reference agents (cimetidine (10 mg, 100 mg, 500 mg/ml), ranitidine (2, 5 mg, 35 mg, 250 mg/ml), famotidine (10 mg, 50 mg, 100 mg/ml), sucralfate (1 mg, 5 mg, 10 mg/ml) either after 3 or 7 days. In other experiments the effect of the pentadecapeptide BPC 157 on NO synthesis was evaluated in vitro, in gastric mucosa from rat stomach tissue homogenates using the same method as before. BPC 157, given in the same dose (100 uM) as L-arginine, induced a comparable generation of NO. But, the BPC 157 effect could not be inhibited by L-NAME, even when L-NAME was given in a tenfold (100 vs. 100 uM)higher dose then that needed for inhibition of the L-arginine effect. NO synthesis was blunted when the pentadecapeptide BPC 157 and L-arginine were combined. In summary, a well known angiogenic effect of sucralfate, we demonstrated an evident angiogenic property of the noven pentadecapeptide BPC 157 could interfere with the NO synthesis in a specific way, especially with L-arginine, having a more prominent and/or particularly different effect from that of NO. Thus, this pentadecapeptide could be of particula importance for the control of endothelium integrity, and processes of healing as well.
