The role of iron in the innate immune system
As a component of many enzymes and oxygen binding carrier proteins, iron is one of the essential micronutrients with a major role in biological systems. Iron availability in the host and its effect on the bacteria growth is one of the best studied phenomena of pathogenicity. The fact that bacterial pathogens need iron for multiplying and achieving full virulence indicates that during the co-evolution with the host they had to adjust to the extracellular environment without iron and develop several mechanisms for obtaining protein-bound host iron. Some of this strategies include modification of iron release from ferritin and utilization in the heme structures, small molecular mass iron chelate production (siderophores), expression of transferrin and lactoferrin receptors, destruction of iron-binding glycoproteins, obstruction of iron binding and reduced transfer of ferric to ferrous iron form. Bacterial growth inhibition by changing the hosts iron metabolism and withholding of nutrients, nutritional immunity, are one of the first lines of defense. Iron sequestration is the most important form of nutritional immunity. The aim of this paper is to give a summarized review of the innate immune defense mechanisms and iron-binding proteins involved in the competition for iron between pathogen and host.
