Ex situ Chronopotentiometric Stripping of Sulphated Polysaccharides κ-, ι-, and λ- carrageenan in Seawater
Sulphated polysaccharides (S-PS) are a “presodium” catalyst of hydrogen evolution on mercury electrode in a buffered solution. Chronopotentiometric stripping (CPS) method detects two peaks named peak H'PS (more negative one) and peak HPS (less negative one) as a result of catalytic hydrogen evolution. It is not possible to detect peaks H directly in seawater. An ex situ procedure of adsorptive transfer (AdT) is applied to transfer adsorbed S-PS from seawater (pH 8.2) to the buffered solution (pH 5.1) where chronopotentiometric stripping and detection of catalytic peaks was possible (Fig.1) Sulphated galactans carrageenans differ in sulphate content and in the presence of the 3, 6- anhydro-D- galactose, which both influence their conformation in water solution. A static mercury drop electrode having drop size 0.40 and 0.52 mm2 was a working electrode. The catalytic peak HPS of κ-, ι-, and λ- carrageenan in seawater was examined at different experimental conditions using AdT procedure coupled with constant current chronopotentiometric stripping and detection. The relation of κ-, ι-, and λ-carrageenan structure and conformation with obtained AdT CPS results was interpreted.
