In vitro regeneration capability of healthy and infected genotypes of cv. Plavac mali

Plavac mali is Croatian autochthonous cultivar of great economic interest. The survey of virus diseases in commercial vineyards of the coastal vine-growing region disclosed a widespread presence of viruses and a low occurrence of non-infected vines in population of this cultivar. In order to start sanitation program, we tested in vitro requirements of healthy and infected genotypes of Plavac mali. Excised nodal segments of healthy genotype were inoculated on three different media: half-strenght MS (M1), full-strenght MS (M2) and full-strenght MS supplemented with 1mg/L-1 BAP growth regulator (M3). Growing rate, evaluated on the base of shoot length (mm) and the number of leaves, were significantly higher on M1 and M2, media without growth regulator, in comparison with M3 medium where callus formation and vitrification were observed. Five genotypes infected with specific combination of viruses (GFLV, GLRaV-1, GLRaV-3, GLRaV-1+GLRaV-3 and GLRaV-3+GVA) were grown on M1 medium. Obtained results showed that sanitary status influenced significantly on regeneration capacity of nodal segments in culture in vitro. Shoot length and the number of leaves of healthy Plavac mali were significantly higher than those of any other infected genotype. Difference in growing rate between genotypes infected with different viruses or virus combination where also observed. The genotype with virus GLRaV-3 showed the lowest results in shoot length and the number of leaves.
