Expression of β-cryptogein stimulates rosmarinic acid secretion from the Coleus blumei transgenic roots

Rosmarinic acid (RA) has been studied intensively for its antimicrobial activity against certain bacteria and fungi. Accumulation of RA in plants is often part of the defense response against stress and pathogen attack, which can be triggered and activated by elicitors. It is well known that oomycetal elicitor, β-cryptogein, induces hypersensitive response and systemic acquired resistance against some pathogens. In order to test the effect of endogenously synthesized β-cryptogein on RA accumulation in tissue, and its secretion into the culture medium, Coleus blumei transgenic roots were established. Agrobacterium rhizogenes was employed to insert synthetic crypt gene, encoding β-cryptogein, under the control of alcoholinducible promoter. The expression of β-cryptogein, in C. blumei hairy roots, was controlled by application of 1% and 2% ethanol, during 21-day-induction period. Ethanol-induced expression of β-cryptogein caused significant decrease of RA in hairy root lines and its increase in culture medium. These data suggest that inducible expression of β-cryptogein might be a potential regulatory factor for secretion of antimicrobials, such as RA, from transgenic tissues.
