Allium x cornutum (2n=3x=24) is a vegetatively propagated triploid hybrid of common onion, Allium cepa L. In an attempt to identify the parental species of that natural hybrid, analyses of several molecular markers (isozymes, RFLP and RAPD) gave poor and somewhat contradictory results. Results of genomic in situ hybridization proved that common onion and A. roylei Stearn could be donors of two of the three genomes of A. x cornutum. Further progress in the identification of the parental species of A. x cornutum was hampered by the lack of more molecular data about wild relatives of common onion. Recently, the ITS sequences (internal transcribed spacer of the 18S-5.8S-26S rDNA) of a large number of common onionrelatives originating from Central Asia, were deposited in GenBank. The aim of this study was to determine the ITS sequences of A. x cornutum and to compare them to other available ITS sequences of the genus Allium L.. For that purpose, the ITS sequences (ITS1, 5.8S rDNA and ITS2) of A x cornutum individuals originating from four localities in Dalmatia (Hvar, Kaštela, Dubrava i Vis) were PCR amplified, cloned and sequenced. The two types of ITS sequences were detected. A smaller number of sequences were of common onion type. Unexpectedly, a much larger number of the ITS sequences were highly similar to those of wild Asian species, Allium pskemense B. Fedtsh, thus indicating that these two species might be progenitors of A. x cornutum. Further analysis will help to clarify the origin and genome structure of that interesting triploid hybrid.
