Influence of persistent infection to molecular diagnostic of equine viral arteritis
In this study four Lipizzaner stallions from the same stud farm were selected. All animals were, two consecutive years, serological positive with VN-test and confirmed as virus shedders by RT-PCRs. Two semen samples were taken from each stallion in two consecutive years. All four semen samples were RT-PCRs positive and RT-PCRs products were sequenced. Results of genetic analysis showed that observed nucleotide substitution in ORF7 EAV from different animals in first samples as a substitution found in ORF7 in stallion 1 in two consecutive years were silent mutations, and amino acids sequence were identical in all eight semen samples. This result confirmed that in generally ORF7 gene is very conservative region of EAV even in persistently infected stallions. Furthermore, despite that many studies had previously shown that genetic and phenotypic variations of EAV is significantly higher in persistently infected stallions results of this study confirmed that RT-PCRs diagnostic based on primers derived from ORF7 gene is trustworthy for diagnostic of EVA even in long time carriers.
