Effect of salt stress on Chinese cabbage (Brassica rapa L.) seedlings measured by spectrophotometric methods
High salt concentrations cause osmotic stress during which generation of reactive oxygen species (ROS) is occured. Plants, like other living organisams, developed antioxidant systems (antioxidant enzymes, tripeptide glutathione and various secondary metabolites with antioxidant activity) whose role is to protect them from unfriendly salt/stress enviromental conditions. During many years, spectrophotometric methods for measuring plants defense mehanisms have been established. The aim of this study was to investigate 1-day old Brassica rapa seedlings respose to abiotic stress using UV/VIS spectroscopy. Abiotic salt stress was induced by NaCl in range of concentrations from 50 mM to 200 mM. The effect of NaCl on seedlings growth and development was determined by using root growth bioassay. Levels of reduced (GSH) and oxidized (GSSG) glutathione, H2O2 and lipid peroxidation were determined as oxidative stress indicators by monitoring apsorbance at specific wavelengths.
