Low dose acarbose effect on the catalitic concentration of maltase in the proximal part of intestine of NOD diabetic mice

The alpha-glucosidase inhibitor acarbose has been shown to competitively inhibit the intestinal digestion of starch as well as sucrose. Addition of acarbose to starch or sucrose load reduces the postprandial glucose and insulin responses in a dose-depended manner. The aim of this study was to determine effectiveness of a low dose of acarbose (25 mg/100 g chow) on the catalytic concentration of intestinal maltase in NOD diabetic mice. Diabetes was induced in NOD mice by i.v. injection of aloxan-monohidrate (75 mg/kg body mass). There were four groups containing seven NOD mice in the experiment: control (K) and diabetic (D) mice feeding with ordinary chow, control (K/A) and diabetic (D/A) mice feeding with the chow prepared with acarbose. They were on this type of diet for seven days. Catalitic concentration of maltase was determined in the homogenate of the proximal part of intestine by the Dalqist method using maltose as substrate. Mean catalytic concentration of maltase was 22 or 25 U/mg proteins in the proximal part of intestine of the control NOD mice regardless of the type of the diet (K and K/A). In the same part of the intestine of two NOD diabetic groups, 32 (D) and 28 (D/A) U/mg proteins were determined, respectively. Acarbose administered in a manner analogous to that used in other animal studies did not show inhibitory effect on the catalytic concentration of maltase in the proximal part of intestine in NOD control or diabetic mice.
