Phycotoxin profile along Eastern Adriatic Coast

National monitoring program of shellfish resources in Croatian waters was established in 2000 including both seawater and shellfish analysis. Samples were collected weekly during warmer part of the year (April-October) and fortnightly from November to March at the breeding areas along Eastern Adriatic coast. Through 2007 and 2008 year 768 shellfish samples were analyzed for Diarrhetic and Paralytic Shellfish Poisoning toxins (DSP, PSP) using mouse bioassay method (MBA), and Amnesic Shellfish Poisoning (ASP) toxins using UV-DAD-HPLC system. Our results highlighted okadaic acid (OA), OA derivates and yessotoxins as major phycotoxins in Croatian shellfish during investigating period. Mouse bioassay for DSP showed 34 positive samples which were further analyzed by LC-MS/MS. These analysis detected yessotoxin and its analogues in 13 samples but in concentration below regulatory limit. OA and its derivates were detected in 5 samples out of which 3 samples were above regulatory limit. When toxins were confirmed in shellfish tissue by LC-MS/MS, analysis of seawater showed presence of several potentially toxic phytoplankton species in abundances not higher than 103 cell m-2. Among them Dinophysis caudata stands out as the most abundant and the most common species. MBA for PSP toxins during the investigating period was negative. Domoic acid (ASP) was detected in 10 shellfish samples in low concentration, below regulatory limit. Seawater analysis confirmed presence of potentially causative phytoplankton species Pseudo-nitzschia spp.
