Distribution of mono-, di- and tri-sialo gangliosides in the brain of Actionopterygian fishes

Mono-, di- and tri-sialo gangliosides are the major glycosphingolipids in the brain of higher vertebrates involved in assembling of lipid rafts. Contrary, fish brain is rich in polisyalo- gangliosides whose function is implicated in modulating repulsive and attractive interactions between cells during embryonic development and process of temperature adaptation. Histological distribution of ganglioside is usually studied in rodent and mammalian brain, but not yet described in fish brain. Gangliosides were extracted from adult brains of trout, carp and zebrafish and separated by TLC. High-affinity anti-ganglioside (GM1, GD1a, GD1b, GT1b) IgG antibodies were used for immunohistochemistry. In trout and carp brain GM1 and GT1b are expressed on the same neuronal cell bodies from telencephalon to spinal cord. In zebrafish brain GM1 was not detected, while GT1b is general neuropil staining. GD1a is specific for unmyelinated parallel fibers in carp and zebrafish brain as well as parallel fibers in molecular layer of all cerebellar divisions. In trout brain GD1b is found on parallel fibers of cerebellum, but not in tectum mesencephali. GD1b is expressed on neuronal cell bodies in zebrafish. Each studied species has different expression of complex gangliosides. GT1b is widely present, while GD1a and GD1b appear on specific group of unmyelinated fibers and could be used as their specific marker. General Significance: This is the first report of mono-, di- and tri-sialo gangliosides (GM1, GD1a, GD1b and GT1b) distribution in the brain of adult Actinopterygian fishes.
