Effect of Pelargonium radula (Cav.) L, Herit on the albumin synthesis in Hep G2 cells under hyperglycaemic conditions
In the present study we measured albumin concentration in a cell culture medium as a marker of albumin synthesis in HepG2 cells. Albumin is secreted rapidly by cells after it is produced. Diabetes mellitus affected the synthesis rates of albumin. In the case of Hep G2 cells, the intracellular retention half-time of albumin is about 35 min. Thus, changes in the concentration of albumin in Hep G2 cells can be assumed to closely reflect changes in albumin synthesis. Six different extracts (E1 – E6) from leaves of the Pelargonium radula (Cav.) L, Herit, cultivated in the Botanical Garden of Faculty of Pharmacy and Biochemistry, Zagreb we prepared by water or alcohol extraction, filtrated and lyophilized. Hep G2 cells were incubated with 20 mM glucose and with different extracts (E1 – E6) in concentration of 25 g/mL. We measured in medium concentration of albumin by Bromcresol green (BCG) Dye Binding Method. Albumin binds quantitatively with BCG at pH 4.15 resulting in the formation of a green colour which can be measured at 630 nm. The results of present study suggested that extract E2 (Soxhlet extraction with 96% ethanol for 2.5 h was applied and then lyophilized) significantlly increase albumin synthesis in Hep G2 cells when compared with cells treated only with glucose (p 0.05).
