Intestinal microbiota of wild chub (Squalius cephalus) infected by Pomphorhynchus laevis (Acanthocephala)
Intestinal bacteria composition of 29 chubs (Squalius cephalus) with different Pomphorhynchus laevis (Acanthocaphala) infection intensity (0 - 95 species) were examined. Chubs were sampled from five locations of Sutla river and gut tissue bacteria were cultivated on Tryptic Soy Agar (TSA). From the TSA plates 82 colonies were randomly collected and purified cultures were obtained for further characterisation. Out of these, 54 isolates, considering different colour and shape, were analyzed by RFLP of PCR amplified 16S rDNA, using HinfI, AluI and RsaI. Bacteria with different restriction profiles were further identified by partial sequencing of the 16S rDNA. From 20 different isolates, 70% of bacteria were members of the class Gammaproteobacteria, 10% of classes Actinobacteria and Bacilli, and 5% of classes Betaproteobacteria and Flavobacteria. Identified Gram-positive bacteria were: Exiguobacterium sp., Staphylococcus aureus, Brachyobacterium paraconglomeratum te Microbacterium sp.. Finding of Aeromonas salmonicida, causative agent of the destructive disease furunculosis proved intestinal tissue as one of the main route for this bacteria spreading. Composition as well as bacterial count in examined chubs intestine were similar in infected and non-infected chubs. Chub species with the most intense P. laevis infection had the most diverse gut bacterial community.
