In vitro immunogenicity of inactivated Parapoxvirus ovis for lymphocytes from domestic animal species and humans

In immunology, the in vitro lymphocyte cell cultures stimulated with various polyclonal stimulators are used as a correlate for the assessment of in vivo cellular immune responsiveness to wast array of microbial/parasitic antigens, antibodies, cytokines, growth factors, nutrients, allogeneic/xenogeneic cells, and cytotoxic and therapeutic agents. The aim of the current study was to establish the capacity of freeze-dried preparation of inactivated Parapoxvirus ovis recommended by manufacturer (Bayer, Leverkusen, Germany) as a new formulation of an immune response modifier (Baypamun-N, BPMN) to induce proliferation of the immune cells from domestic animal species (rabbit, goat, swine, cow, horse, dog, and cat) and humans. Blood samples were collected into heparinized syringes and PBL were cultured with or without mitogens (PHA, ConA, PWM)/specific antigens (LPS, KLH, BPMN). All three concentrations of BPMN induced comparable proliferative responses of PBL from species tested to those induced by either mitogens or specific antigens. However, concentration of 20 ľl/ml of the preparation stimulated goat, rabbit, and cat PBL much better than any of stimulators applied. BPMN at this concentration stimulated PBL from horse, dog, pig, and man to respond with at least 50% of the average stimulation index (SI) value. At 10 ľl/ml BPMN highly stimulated caprine, equine, and human PBL (>100% of average SI value), and moderately (>50%) the cells from cat and pig. The lowest concentration of BPMN tested (5 ľl/ml) induced high response of bovine, porcine, and human immune cells (>100%), and good response of equine PBL (>95%). Generally, BPMN exhibited strongest stimulatory effect for porcine, caprine and human lymphocytes, moderately high effect for horse, cat, cow, and rabbit, and rather low effect for dog PBL.
