INTERACTION OF TONALIDE WITH CYP1A AND ABC TRANSPORT PROTEINS IN FISH CELL LINE PLHC-1
The polycyclic musk Tonalide (AHTN) is one of most common synthetic musk fragrances used in household and personal care products. The continuous release and bioaccumulation potential in aquatic environment underline the importance of investigating toxicological targets. Few data are currently available on both access and cellular metabolism in aquatic organisms. The aim of the present study is to investigate the interaction of AHTN with CYP1A and ABC transport proteins using PLHC-1 fish cell line. Cyp1a gene transcription and related EROD activity were measured as well as abcc2-3 (encoding for Multidrug Resistance Associated Proteins, MRPs) and abcb1 (P-glycoprotein ; P-gp) genes. Three doses of AHTN (1nM, 10nM and 2 µM) for 6 and 24 h were tested in single and co-exposure with known CYP1A inducers as B(a)P and PCB 126. Only a slight induction of cyp1a gene transcription was observed after 6 h compared to controls (2.3 folds at 2µM). At 24 h, no differences were observed. Both co-exposure with AHTN caused a strong decline of cyp1a transcription levels compared single inducers exposure at 6 h. EROD activity was not affected after 6 and 24 h of exposure to AHTN and also in co-exposure with PCB 126. Concerning ABC transporters, at 6 h only abcc3 was slightly induced (about 2 folds vs ctrl) while no effects on the other two genes, abcb1 and abcc2 were observed. At 24 h, a general reduction of transcription of all three transporters was observed. Results indicate that AHTN is not metabolized by CYP1A, nevertheless the compound may be actively metabolized and excreted from the cell within 24 h probably by MRP3. AHTN is able to reduce detoxification capacity of CYP1A with consequent increased toxicity of other contaminants.
