Effect of plant mixture extract P-9801091 on the concentration of fructosamine, pyruvate kinase and ATP-citrate lyase in diabetic NOD mice.
The aim of this study was to assess changes in the concentrations of fructosamine in serum and pyruvate kinase (PK) and ATP-citrate lyase (citrate cleavage enzyme, CCE) in the liver of diabetic NOD mice treated with plant mixture extract P-9801091 (PME). Fructosamine is a generic name for plasma protein ketoamines. The concentration of glycated albumin reflect the control of glucose during the preceding 2 to 3 weeks. We chose pyruvate kinase (PK) as glycolytic enzyme and ATP-citrate lyase (CCE) as lipogenic enzyme to estimate the effect of PME on the carbohydrate metabolism in NOD mice. Diabetes mellitus in NOD mice was induced by i.v. injection of alloxan in dose of 65 mg/kg b. w. The treatment with PME started 7 days after alloxan application. PME in dose, 20 mg/kg b.w.,was admixed to food. NOD mice took food with PME during 7 days before enzyme activities determination. The plant mixture extract P-9801091 (Zagreb, Croatia) was obtained by ethanol extraction followed by alcohol evaporation and lyophylisation. Results showed that fructosamine level in serum of diabetic NOD mice (347.4ĄÓ10.7 Ýmol/L) statisticaly increase (p<0.01) compared to healthy control mice (219.8ĄÓ6.0 Ýmol/L). After 7 days treatment with PME, fructosamine level in the serum of diabetic mice (210.2ĄÓ3.4 Ýmol/L) statisticaly decreased and almost reach the healthy control values. The activity of PK statisticaly (p<0.01) decreased, about 80 % (10.0b3.8 nmol/mg prot./min) compared to healthy controls (48.6b8.0 nmol/mg prot./min). Treatment with PME increased PK activity about 30 % in the liver of diabetic mice (32.0b10.1 nmol/mg prot./min). CCE activity statisticaly (p<0.01) decreased, about 70 % (4.2b0.6 nmol/mg prot./min) in diabetic group compared to healthy mice (14,7b1.3 nmol/mg prot./min). After PME treatment values decreased about 20 % (7.6b2.2 nmol/mg prot./min) according to non-treated diabetic mice. We can conclude that PME (P-9801091) applied 7 days as a food aduvant can almost restore fructosamine level in serum of diabetic NOD mice. Liver carbohydrate metabolisam of diabetic mice was restored between 20ĄV30% after 7 days of PME use.
