Competitive exclusion and immunomodulation mediated by S-layer protein from Lactobacillus helveticus M92

Lactobacillus helveticus M92, besides health promoting effects as probiotic, has also a potential as a carrier for the oral and mucosal vaccines due to the presence of S-layer proteins. The aim of this study was characterisation of the functional role of S-layer from L. helveticus M92 in the immunomodulation and competitive exclusion of Salmonella enterica serovar Typhimurium FP1 exerted by L. helveticus M92 application in mice. S-layer protein was identified by SDS-PAGE LC-MS/MS analysis. Sequence similarity searches of S-layer protein from L. helveticus M92 revealed high homology with other well characterised S-layer proteins of Lactobacillus strains. Purified S-layer protein has shown significantly better immunomodulatory effect in orally immunised mice than L. helveticus M92 cells after S-layer protein removal. S-layer was found to be involved in the autoaggregation of L. helveticus M92 cells, coaggregation of L. helveticus M92 with S. Typhimurium FP1 and adhesion to Caco-2 cells, as these processes were negatively affected after S-layer proteins removal from the cell surface. Therefore, the influence probiotic strain L. helveticus M92 on an oral infection of mice by S. Typhimurium FP1 was investigated. Following the oral immunization of mice, with viable L. helveticus M92 and S. Typhimurium FP1 cells, the concentration of luminal content of total S-IgA and specific anti-Salmonella S-IgA antibodies, from all immunized mice was significantly higher compared to the control group or group of mice infected only with S. Typhimurium FP1. These results demonstrate that reduced infection by S. Typhimurium FP1 in mice, when probiotic strain was applied, is associated competitive exclusion of S. Typhimurium FP1 in the intestinal tract and enhanced immune protection conferred by the L. helveticus M92 and its S-layer protein.
