Superoxide dismutase in leukocytes and erythrocytes of children with asthma

Background and purpose: In exacerbation of asthma a strong inflammation is developed and inflammatory cells become activated. Due to this activation, the respiratory burst occurs in neutrophils, eosinophils, monocytes and macrophages. Oxygen radicals produced by involved cells are held responsible for damaging airway's epithelia and endothelia, causing mucus hypersecretion and increase in vascular permeability with formation of edema and bronchoconstriction. Superoxide dismutase exhibits protection from oxidative damage by dismutation of superoxide radicals. Materials and methods: Activity of superoxide dismutase was measured spectrophotometrically in erythrocytes and leukocytes from healthy adults (control group) and from patients, children with chronic asthma. The analysis was performed at two times on the same patient: in acute period of disease (group 1) and in the asthma-free period (group 2). Patients were under therapy with salbutamol. Interference of salbutamol and teophilin with analytical method for determination of superoxide dismutase was examined. Results: Activity of superoxide dismutase in leukocytes decreased in group 1 compared with group 2 for 31% (p<0.05), and for 36% compared with control group (p<0.05). Superoxide dismutase activities in leukocytes from group 2 and from control group did not differ significantly. Differences in activities of superoxide dismutase from erythrocytes were not significant. No important analytical influence of salbutamol was found. Conclusions: The superoxide dismutase in leukocytes was decreased by exacerbation of asthma and normalised in asthma-free period. Acute asthma did not influence the superoxide dismutase activity in erythrocytes. The impact of antiasthmatic therapy on superoxide dismutase determination was negligible.
